Spurs' manager Juande Ramos had targeted Milito all summer but was put off by the £15million demanded by his Spanish club Real Zaragoza.
But Spurs were offered Milito, 29, for a fraction of that price in the countdown to Monday's deadline and Ramos was desperate to sign him as Dimitar Berbatov headed to Manchester United.
However Comolli was accused of botching the move and Milito opted to join Italian club Genoa despite them offering significantly less than Tottenham's £40,000-a-week wages.
Deteriorated Spurs' chairman Daniel Levy is now considering Comolli's position, with the Frenchman's relationship with Ramos having deteriorated.
Several of Comolli's buys have failed to make the grade since he replaced Frank Arnesen in the job three years ago.
Spurs were also pursuing a second target from Zaragoza, forward Sergio Garcia, who instead signed for Real Betis on Monday. A renewed bid for Atletico Madrid striker Diego Forlan was snubbed by the Spanish club after they qualified for the Champions League group stages.
After the departures of Berbatov, Robbie Keane and Jermain Defoe, Spurs have only two senior strikers on their staff - Russian new boy Roman Pavlyuchenko and Darren Bent, a player Ramos has been sceptical about.
Youngster Fraizer Campbell is on loan from Manchester United, who are unlikely to allow the player to leave permanently if he is a success at Spurs.
